Methods of virus elimination ( or Production of disease-free plants)

  1. Heat treatment:
  • Hot water: Eliminates virus at dormant buds.
  • Hot air: In actively growing plants (35-400C).

 

  1. Meristem Culture:
  • Larger the explant greater the chances of plant regeneration and virus particle presence.

 

 

Why meristem culture?

  • Rate of cell division is higher than virus multiplication.
  • No vascular cells, no movement of virus.
  • High metabolic activity so no virus replication.
  • Auxin level high, inhibit virus multiplication.

 

  1. Chemical treatment:
  • Low success.
  • Includes treatment with malachite green, cycloheximide, actinomycin.

 

  1. Callus Culture

  3. Low heat treatment (4-90C for several months)
  4. Somatic hybridization
