Methods of virus elimination ( or Production of disease-free plants)
- Heat treatment:
- Hot water: Eliminates virus at dormant buds.
- Hot air: In actively growing plants (35-400C).
- Meristem Culture:
- Larger the explant greater the chances of plant regeneration and virus particle presence.
Why meristem culture?
- Rate of cell division is higher than virus multiplication.
- No vascular cells, no movement of virus.
- High metabolic activity so no virus replication.
- Auxin level high, inhibit virus multiplication.
- Chemical treatment:
- Low success.
- Includes treatment with malachite green, cycloheximide, actinomycin.
- Callus Culture
- Transformation
- Low heat treatment (4-90C for several months)
- Somatic hybridization
- Somaclonal variation.
