Agrobacterium tumefaciens mediated transformation:

  • T-DNA is removed from bacterium
  • Plasmid is cut using a restriction enzyme
  • Foreign DNA cut with same restriction enzyme and inserted into plasmid.
  • Formation of recombinant plasmid takes place
  • Recombinant Ti plasmid reinserted into bacterium
  • T-DNA carrying inserted gene incorporated into plant cell chromosome
  • Plant cell is then cultured
  • A transgenic plant expressing inserted gene is then generated.
